The collision, between a van and car, occurred at the intersection of Ventnor and Red Rocks roads about 6.45pm.
Sadly, the male passenger from the car died at the scene.
A woman from the same vehicle had been airlifted to a Melbourne hospital.
Two people from the van, believed aged in their 20s, appeared to have escaped serious injury.
Major Collison Investigation Unit detectives were on scene and were investigating the circumstances surrounding the collision.
